摘要
DNA疫苗是治疗人类与动物疾病的一个迅速发展的下一代疫苗平台。许多DNA投递装置如电穿孔和无针注射器被用来增强基因转染。这也导致了更高的抗原表达与人类-动物的体液和细胞免疫改进的关联。该综述重点介绍了近些年可以改进DNA疫苗性能的载体、转基因的设计创新。这些新的载体可以增强抗原表达,增加生物反应器中的质粒产量与质量,消除抗生素的选择及其它潜在安全问题。我们提出了一个流程图,包括设计合成抗原转基因、结合抗原定位、密码子优化及生物信息学的内容。目前在传染病、癌症、自身免疫、免疫耐受性及过敏适应症的发展中,为确保基因疫苗生产的安全性、有效性、经济适用性,关键是改进的载体、不含抗生素的质粒产品及具有成本效率的制造工艺的应用。

摘要: DNA vaccines are a rapidly deployed next generation vaccination platform for treatment of human and animal disease. DNA delivery devices, such as electroporation and needle free jet injectors, are used to increase gene transfer. This results in higher antigen expression which correlates with improved humoral and cellular immunity in humans and animals. This review highlights recent vector and transgene design innovations that improve DNA vaccine performance. These new vectors improve antigen expression, increase plasmid manufacturing yield and quality in bioreactors, and eliminate antibiotic selection and other potential safety issues. A flowchart for designing synthetic antigen transgenes, combining antigen targeting, codon-optimization and bioinformatics, is presented. Application of improved vectors, of antibiotic free plasmid production, and cost effective manufacturing technologies will be critical to ensure safety, efficacy, and economically viable manufacturing of DNA vaccines currently under development for infectious disease, cancer, autoimmunity, immunotolerance and allergy indications.
