目前传统上的抗病毒药物开发主要是针对疱疹病毒 、人类免疫缺陷性病毒(HIV)和丙型肝炎病毒(HCV),埃博拉病毒,作为一种新的病毒靶点近期已经被广泛关注。对抗病毒药物的十种核苷类似物(或他们的类别)的潜在治疗效果进行了总结:(i) BCX4430一种碳苷;(ii) 4’-叠氮-、4’-氰基-和4’-乙炔基衍生物;(iii) 4’-核苷; (iv) 虫草素 (3’- 脱氧腺苷); (v) 吡唑霉素, 另一种碳苷(vi) neplanocin A 类似物; (vii) EICAR, 一种三唑核苷类似物; (viii) GR-92938X, 一种双甲酰胺; (ix) sofosbuvir (Solvaldi®), 一种2’-C-甲基核苷类似物; 以及(x) 法匹拉韦(T-705), 一种吡嗪类似物。

摘要: In the current context of antiviral drug development, which has been traditionally dominated by herpesviruses, human immunodeficiency virus (HIV) and hepatitis C virus (HCV), a new viral target has been recently gained unforeseen attention, Ebola virus. Ten nucleoside analogues, or categories thereof, are reviewed for their therapeutic potential as antiviral drugs: (i) BCX4430, a C-nucleoside; (ii) 4’-azido-, 4’-cyano-, and 4’-ethynyl derivatives; (iii) 4’-thionucleosides; (iv) cordycepin (3’-deoxyadeosine); (v) pyrazofurin, another C-nucleoside; (vi) neplanocin A analogues; (vii) EICAR, a ribavirin analogue; (viii) GR-92938X, a double carboxamide; (ix) sofosbuvir (Solvaldi®), a 2’-C-methylnucleoside; and (x) favipiravir (T-705), a pyrazine analogue.
